Summary

VK2735 is a dual GIP and GLP-1 receptor agonist developed by Viking Therapeutics, currently in Phase 2 clinical trials for weight management. It is administered as a weekly subcutaneous injection. Early-phase data suggest substantial weight loss, positioning it as a potential next-generation incretin therapy. Research use only — not a licensed medicine.

Mechanism

VK2735 is a dual agonist of the GIP (glucose-dependent insulinotropic polypeptide) and GLP-1 (glucagon-like peptide-1) receptors. GIP receptor activation enhances insulin secretion and may improve adipose tissue metabolism, while GLP-1 receptor activation promotes insulin secretion, suppresses glucagon, delays gastric emptying, and reduces appetite. Combined activation of both receptors is thought to produce additive or synergistic effects on weight loss and glycaemic control, as demonstrated by the success of tirzepatide in this mechanistic class.

Evidence base

VK2735 has completed Phase 2 clinical trials, with the most recent published evidence being the VENTURE study (PMID 41508550) — a Phase 2, randomised, 13-week trial of weekly subcutaneous VK2735 for weight management. Earlier Phase 2 results were reported via press release and conference presentation. The compound is advancing to Phase 3. Evidence is graded as moderate: randomised controlled Phase 2 data exists, but Phase 3 confirmatory trials are ongoing.

Protocols

VK2735 is administered as a weekly subcutaneous injection. Phase 2 trials evaluated multiple dose levels over a 13-week treatment period. Specific dose ranges are being optimised in ongoing studies. No protocols are established outside clinical trial contexts.

VK2735 is an investigational compound not licensed as a medicine in the UK. It is not a controlled substance. It falls into a UK grey area: legal for research purposes but not MHRA-approved for human therapeutic use. Sale for human consumption is illegal under UK medicines regulations.

Vendor notes

VK2735 is an investigational compound in active clinical development by Viking Therapeutics. It is not commercially available through UK peptide vendors. No vetted vendors offer this compound. Caution is advised regarding any supplier claiming to sell VK2735.
